綠茶萃取物EGCG

美國婦產科學期刊研究指出,綠茶萃取物EGCG或許能有效治療影響25%至30%女性的子宮肌瘤。子宮肌瘤會讓人虛弱,造成陰道大量出血、貧血、以及疲勞等。田納西州Meharry醫學院的Dong Zhang醫師與同事們表示,他們發現,綠茶萃取物在實驗室內能殺死子宮肌瘤細胞,讓老鼠的肌瘤縮小並減少重量。

他們的研究結果刊載在美國婦產科學期刊(American Journal of Obstetrics and Gynecology)中。研究人員們將20隻老鼠注射肌瘤細胞,讓其中10隻的飲水混合EGCG,另外10隻喝開水。

EGCG是綠茶中的多酚類。研究時間為8週,在治療4週與8週後,肌瘤長得比較小且重量減少。研究人員指出,在EGCG組中有1隻老鼠在8週後腫瘤消失。

在實驗室中,齧齒類的肌瘤細胞用EGCG治療48至72小時後生長較慢且變小,EGCG也促使肌瘤細胞死亡。研究人員指出,這給他們很大的鼓勵,在他們的研究中,在飲水中添加相當適度劑量的EGCG,於治療8週後能成功誘發肌瘤細胞的大小顯著持續地縮小。

他們的研究指出,長期使用EGCG或許能幫助婦女抑止腫瘤發展,避免大範圍手術的風險。

Green tea extract inhibits proliferation of uterine leiomyoma cells in vitro and in nude mice.

Abstract

OBJECTIVE: The purpose of this study was to investigate the effect of epigallocatechin gallate (EGCG) on rat leiomyoma (ELT3) cells in vitro and in a nude mice model.

STUDY DESIGN: ELT3 cells were treated with various concentrations of EGCG. Cell proliferation, proliferation cell nuclear antigen (PCNA), and cyclin-dependent kinase 4 (Cdk4) protein levels were evaluated. ELT3 cells were inoculated subcutaneously in female athymic nude mice. Animals were fed 1.25 mg EGCG (in drinking water)/mouse/day. Tumors were collected and evaluated at 4 and 8 weeks after the treatment.

RESULTS

Inhibitory effect of EGCG (200 micromol/L) on ELT3 cells was observed after 24 hours of treatment (P < .05). At > or = 50 micromol/L, EGCG significantly decreased PCNA and Cdk4 protein levels (P < .05). In vivo, EGCG treatment dramatically reduced the volume and weight of tumors at 4 and 8 weeks after the treatment (P < .05). The PCNA and Cdk4 protein levels were significantly reduced in the EGCG-treated group (P < .05).

CONCLUSION: EGCG effectively inhibits proliferation and induces apoptosis in rat ELT3 uterine leiomyoma cells in vitro and in vivo.

 

薑黃素Curcumin

薑黃素對子宮肌瘤細胞增殖的抑制作用。子宮肌瘤是最常見的婦科良性腫瘤,它是絕經前婦女中子宮切除的主要原因。薑黃素是薑黃的有效成分,已經報導其預防各種疾病例如癌症,糖尿病和肥胖症。以前的研究報告薑黃素抑制幾種腫瘤細胞的增殖。然而,還沒有精確的表徵薑黃素誘導的子宮肌瘤細胞的抑制作用。在這項研究中,我們調查薑黃素對子宮肌瘤細胞增殖的抑制作用。研究設計:使用Eker大鼠衍生的子宮肌瘤細胞系(ELT-3細胞)。通過計數細胞數和MTS測定來評估細胞增殖。

通過熒光素酶測定評價過氧化物酶體增殖物激活受體-γ(PPARγ)的活化。結果:我們發現薑黃素顯著抑制ELT-3細胞增殖。 PPARγ在ELT-3細胞中表達,薑黃素作為PPARγ配體。薑黃素的這種抑製作用通過用PPARγ拮抗劑處理細胞而減弱。

結論:體外實驗結果顯示,薑黃素對ELT-3細胞增殖的抑製作用通過PPARγ的激活而發生。薑黃素可用作子宮肌瘤的替代療法。

Inhibitory effect of curcumin on uterine leiomyoma cell proliferation.

Abstract

OBJECTIVE: Uterine leiomyomas are the most common gynaecological benign tumour and greatly affect reproductive health and wellbeing. They are the predominant indication for hysterectomy in premenopausal women. Curcumin, a well-known component of turmeric, has been reported to prevent various diseases such as cancer, diabetes and obesity. Previous study reported that curcumin represses the proliferation of several tumour cells. However, there has not been a precise characterisation of the curcumin-induced inhibition of uterine leiomyoma cells. In this study, we investigated the inhibitory effect of curcumin on leiomyoma cells proliferation.

STUDY DESIGN: Eker rat-derived uterine leiomyoma cell lines (ELT-3 cells) were used. Cell proliferation was assessed by counting the number of cells and MTS assay. The activation of peroxisome proliferator-activated receptor-gamma (PPARγ) was evaluated by luciferase assay.

RESULTS

We found that curcumin significantly inhibited ELT-3 cell proliferation. PPARγ was expressed in ELT-3 cells and curcumin acted as a PPARγ ligand. This inhibitory effect of curcumin was attenuated by the treatment of cells with PPARγ antagonist.

CONCLUSION: These experimental findings in vitro show that the inhibitory effect of curcumin on ELT-3 cell proliferation occurs through the activation of PPARγ. Curcumin may be useful as an alternative therapy for uterine leiomyoma.
